When you start a game full screen. Will the game have all available memory.
So for example you have a 256mb card. When running a game will the game have all 256mb of ram. Or will the desktop since it uses 3D be using a portion of that 256mb of ram

Essentially yeh. Windows will recognise that certain desktop features won't be needed or used, so it'll take away the Glass (essentially, bit more in-depth than that) and depending on your driver settings (in Device Manager) and the game settings, it should take full advantage of your card :o)
Even if it doesn't right now (depending on the build of Vista you have), it should do by the end product.
